Pesquisadores da Universidade Estadual Paulista (Unesp) sintetizaram um novo composto que inibe a replicação do vírus da hepatite C em diversos estágios de seu ciclo. O estudo, feito com apoio da Fundação de Amparo à Pesquisa do Estado de São Paulo (Fapesp), consistiu na combinação de moléculas já existentes para produzir novos compostos biológicos, método denominado bioconjugação. O novo composto, denominado AG-hecate, também é capaz de agir em bactérias, fungos e células cancerosas.

Segundo um dos autores do estudo, o químico Paulo Ricardo da Silva Sanches, esse tipo de atuação não é comum nos antivirais que normalmente têm alvos específicos isolados, inibindo processos específicos como a entrada do vírus nas células, a síntese do material genético e de proteínas, a montagem e liberação de novas partículas virais.

“O AG-hecate, ao contrário, apresentou ampla atividade, agindo em diversas etapas do ciclo. O composto também apresentou atividade nos chamados ‘lipid droplets’ – gotas de lipídeo no interior das quais o vírus circula nas células e que o protegem do ataque de enzimas. O AG-hecate desestrutura essas gotas de lipídeo e deixa o complexo replicativo do vírus exposto à ação das enzimas celulares”, disse o químico.

Molécula

De acordo com o estudo, o composto também mostrou alto índice de seletividade, o que significa que ele ataca muito mais o vírus do que a célula hospedeira, mostrando potencial para atacar a doença. “Apesar do composto apresentar pequena atividade nos glóbulos vermelhos do sangue, a molécula precisa passar por alterações em sua estrutura para reduzir ainda mais a sua toxicidade. É nisso que estamos trabalhando agora, para que a pesquisa possa evoluir da fase in vitro para a fase in vivo”, afirmou Paulo Ricardo.

O estudo e o desenvolvimento da molécula AG-hecate demorou cerca de dois anos. Até que entre no mercado e passe a ser utilizada serão necessários mais oito anos, já que o tempo médio para planejamento e desenvolvimento desse tipo de medicamento é de dez anos.

Para o professor Eduardo Maffud Cilli, orientador do estudo, a molécula também age em bactérias, fungos e células cancerosas. No caso do câncer, a molécula interage e destrói a membrana da célula afetada. “Além disso, como os vírus do zika e da febre-amarela apresentam ciclos replicativos bastante parecidos com o do HCV, vamos testar a efetividade do AG-hecate também em relação a esses vírus”, concluiu.
